The light reaction of photosynthesis is very important. Light energy absorbed by pigments in the chloroplast (see Week 1 Lab) is rapidly and efficiently converted into ATP and a reduced molecule, NADPH. During this conversion, the energy from light is briefly converted into an excited electron that is passed from protein to protein in the thylakoid membrane. In this lab we will "eavesdrop" on the process to examine how effectively different wavelengths of light produce excited electrons in the light reaction.

You will now measure the photosynthetic activity of your chloroplasts using a reaction discovered by Robert Hill in 1937. The Hill reaction uses a molecule called DCIP (2,6-dichloroplenolindophenol). DCIP in it's oxidized form is blue. In the presence of chloroplasts, DCIP will steal electrons from the light reaction. Specifically, the electrons are accepted by DCIP just before they reach photosystem I (PSI). (Look at the image of the light reaction in the Introduction of this lab to identify where PSI is in the light reaction pathway.) When DCIP becomes reduced by these electrons it changes from blue to clear…and you can watch this change happening using the spectrophotometer.
